What is TB-500?
TB-500 is a synthetic peptide corresponding to the actin-binding region of Thymosin Beta-4 (Tb4), a naturally occurring 43-amino-acid protein found at high concentrations in blood platelets and wound fluid. The active sequence LKKTETQ within Tb4 is responsible for its G-actin sequestration activity, which underlies many of its biological effects on cell motility, differentiation, and tissue remodeling.
Research Context
TB-500's central mechanism — sequestration of G-actin monomers — prevents their polymerization into F-actin filaments, fundamentally altering cytoskeletal dynamics and promoting cell migration. Studies have documented enhanced keratinocyte, endothelial, and fibroblast migration in wound models, along with accelerated angiogenesis and vasculogenesis.
